The PAXgene Blood ccfDNA products are part of a streamlined system for the collection of whole blood that stabilizes the concentration of circulating cell-free DNA (ccfDNA) in plasma and enables the subsequent purification of ccfDNA for sensitive downstream applications.
The PAXgene Blood ccfDNA System is for Research Use Only. Not for use in diagnostic procedures. The system consists of a blood collection tube (PAXgene Blood ccfDNA Tube) and a ccfDNA purification kit for the QIAGEN QIAsymphony SP instrument (QIAsymphony PAXgene Blood ccfDNA Kit). The system is intended for the collection, storage and transport of blood, the stabilization of ccfDNA in a closed tube, and the subsequent isolation and purification of ccfDNA from plasma for research applications.
Different anticoagulants (EDTA, citrate or heparin) and crosslinking reagents in blood collection tubes can cause variation in the quality and integrity of circulating, cell-free DNA (ccfDNA) isolated from whole blood specimens. PAXgene Blood ccfDNA Tubes contain a reagent that stabilizes ccfDNA levels in plasma without impacting downstream assays.
Apoptosis and lysis of white blood cells in unstabilized whole blood releases intracellular DNA into plasma, which dilutes naturally occurring ccfDNA. The PAXgene Blood ccfDNA Tubes contain an additive that anticoagulates blood and stabilizes blood cells via a non-crosslinking stabilization solution, thereby preventing cell apoptosis. This helps block the release of intracellular DNA and stabilizes ccfDNA levels in plasma.
PAXgene Blood ccfDNA Tubes filled with whole blood can be used directly for the isolation of ccfDNA using a dedicated single-tube handling protocol for automated purification with the QIAsymphony PAXgene Blood ccfDNA Kit. Purification can also be done manually using the QIAGEN QIAamp Circulating Nucleic Acid Kit.
