Traditional tissue fixation methods have been of limited use for molecular analysis. Formalin introduces crosslinks between biomolecules, which leads to degradation and chemical modification of nucleic acids. Stabilization methods for preservation of nucleic acids in tissue, do not preserve histomorphology. To enable both molecular and traditional pathology testing from the same specimen, a method is needed for stabilization of molecular content and preservation of morphology.
To meet such needs PreAnalytiX offers a system for simultaneous stabilization of molecular content and preservation of morphology. Without crosslinking, the PAXgene Tissue System enables histological analysis and extraction of high-quality RNA, miRNA, and DNA from the same specimen.
PAXgene Tissue Container for Tissue Fixation and Stabilization
The PAXgene Tissue Container is designed for standardized collection, stabilization, storage, and transportation of human tissue specimens. The prefilled dual-chambered container contains PAXgene Tissue Fix for rapid penetration and fixation and PAXgene Tissue Stabilizer for stabilization of biomolecules and histomorphology. Stabilized samples can be embedded in paraffin for histological studies. Nucleic acids can be isolated from the stabilized samples using PAXgene Tissue Kits before or after embedding in paraffin.
Histomorphology Preservation
After processing and paraffin embedding, sections can be used for all kinds of traditional as well as immuno staining methods. Equivalent histological details but a clearer chromatin structure and more nuclear details in H&E staining are achieved.
Biomolecule Preservation
For purification of nucleic acids, the system requires the use of one of the PAXgene Tissue Kits for RNA, miRNA, or DNA. Purification of RNA, miRNA, and DNA can be performed from one sample.
PAXgene Tissue Kits provide intact nucleic acids without crosslinking for improved molecular results and sensitive downstream applications.
